Abstract : The emerging concept of Cancer Stem Cells (CSCs) has been of growing interest in recent years. However, the contribution of CSCs to malignant progression of tumors and the role they play, especially in low grade tumors, remain under investigation. In Meningiomas, tumors that occur in multiple extra-axial locations within the arachnoid membranes, meningioma cancer stem cells (MCSCs) have been identified, however, their full identity, in situ spatial distribution, heterogeneity, and the relationship with tumor grade, remain elusive. In this study, we aimed to determine the heterogeneous characteristics of MCSCs in situ, characterize cancer stem cells component in meningiomas and determine which CSC biomarkers are highly expressed in meningioma tissues. Double immunofluorescent-staining was performed on eight frozen meningioma tissues stained with anti-Nestin, anti-Ki67, anti-CD133, anti-SOX2, anti-Vimentin, anti-Frizzled 9, anti-GFAP, anti-beta III Tubulin, anti-SSEA4, anti-SOX2, anti-SSEA4, and anti-Olig2. Statistical analysis was carried out using Microsoft Excel 2010. We observed the patterns of expression for all biomarkers investigated in all tumors. Expression levels of Ki67, SOX2, and Frizzled9 were low whereas levels of Vimentin, SSEA4, and Olig2 were mostly high. Expression levels of Nestin, CD133, GFAP, and BIII were variable across all tumors. Furthermore, correlation analysis showed significant relationships between the expressions of Ki67 with Nestin, Vimentin and Frizzled 9. Cells positive for double staining of CD133/SOX2, and Vimentin/Frizzled9 had low levels of expression, whereas various levels were observed for double stained cells of SSEA4/Olig2 in all tumors. Our work supports the notion of a heterogeneous population of CSCs meningiomas, even in low grade tumors, and calls for the use of more practical clinical diagnostic protocols that can quantitate CSCs, predict tumor recurrence, assist in guiding treatment selection for inoperable tumors, and improve follow up of therapy.
