Abstract : During inflammation, neutrophils are rapidly recruited at sites of acute infection and dominate the initial influx of leukocytes. Two intracellular signalling pathways have been found to be of particular importance in the recruitment, activation and survival of neutrophils: phosphoinositide 3-kinase (PI3K) and p38 mitogen-activated protein kinas (p38 MAPK). This study aimed to investigate the impact of PI3Kγ inhibitor on intracellular signaling enzymes as respiratory burst responses . The study included 10 normal, healthy volunteers (age range: 15-50 years) not suffering from any systematic disease or taken any medication. Blood was collected on heparinzed tube then the neutrophils were isolated and counted. Superoxide anion generation was measured as the superoxide dismutase (SOD) inhibited reduction of ferricytochrome c. The enzyme linked immunosorbent assay system (ELISA) was used to read the different concentrations of RANTES and PI3Kγ II inhibitors. Results obtained showed that, the optimal concentration of RANTES-induced superoxide production from human peripheral blood neutrophils was 10-7μM and the optimal concentration of PI3Kγ II inhibitors which can inhibit O2- generation was 10-5μM. The comparison between the controls and the different concentrations of RANTES, PI3Kγ II inhibitors with or without SOD by using t-test showed that there are no significant differences between neither the negative nor the positive controls and the different concentrations of stimuli or inhibitor with or without SOD. In conclusion, RANTES-induced the respiratory burst in human peripheral blood neutrophils that inhibited partially by PI3Kγ inhibitor II.
