Abstract : Plant growth-promoting bacteria (PGPB) and silicon (Si) can augment salt stress tolerance in plants. In this study, 25 potential PGPB strains were isolated from alfalfa rhizosphere and screened in vitro for their ability to synthesize indole-3-acetic acid, 1-aminocyclopropane-1-carboxylate deaminase, and solubilize tricalcium phosphate. Two promising strains were tentatively identified as Pseudomonas pseudoalcaligenes (KB-10) and P. putida (KB-25) based on their phenotypic, biochemical and 16S rRNA gene phylogeny. Subsequently, a pot experiment was carried out to evaluate the effectiveness of KB-10 and KB-25 treatment, alone or in combination with foliar spray of Si, in alleviating salt stress in coriander. The results showed that salt stress significantly decreased relative water content (RWC), concentrations of photosynthetic pigments (chlorophyll a, chlorophyll b, total chlorophyll and carotenoids), peroxidase (POD) activity, total biomass, and increased total phenolic contents (TPC) in coriander. On the other hand, treatment with PGPB strains and/or Si significantly increased RWC, photosynthetic pigment concentrations, POD activity, total biomass, salt tolerance index, and reduced salt-induced TPC. Overall data showed that plants treated with KB-10 and Si followed by KB-25 and Si performed better in terms of aforementioned parameters as compared to plants treated with KB-10 or KB-25 or Si alone under both non-saline and saline conditions. These results indicate that the combined application of PGPB and Si could be a feasible and effective approach to improve growth and salt stress tolerance in coriander. To the best of our knowledge, this is the first report on PGPB and/or Si mediated salt stress tolerance in coriander.
