Abstract : To investigate pulpal reaction to the EndoSequence Root Repair materials and Low-level laser application in terms of reparative dentin formation and inflammatory changes using histological stains and immunohistochemical analysis. Material and Methods: Eighty teeth in 8 Canaan dogs were randomly assigned to four main groups (n=20). After exposing the pulp via class V, half of the samples received low-level diode laser at 870 nm with dose of 3.24 J/cm2. Thereafter, cavities were capped with fast-set or regular-set EndoSequence Root Repair Material. Animals were scarified after two weeks and two months. Specimens were processed for histomorphological and immunohistochemical examination. Calcific barrier formation and pulpal status were evaluated. Statistical analysis was performed using Kruskal-Wallis and Mann-Whitney U tests. P value was set significant at 0.05. Results: Complete dentin bridges were formed in 84% of samples. Low-level laser group had significantly more reparative dentin area than Non-lased group (P < .05). Low-level laser group exhibited more angiogenesis at 2 weeks. Tubular pattern of calcific barriers was found significantly at 2 months compared to 2 weeks (P <.05). Double bridging was associated with regular-set more than fast-set EndoSequence Root Repair Material. Initial mild to moderate inflammation among groups that subsided significantly after 2 months (P < .05). Runx2 expression was remarkable in Low-level laser group compared to non-lased at 2 weeks. Osteocalcin staining was more at 2 months than at 2 weeks. Generally, intensity of Runx2 expression was greater than osteocalcin. Conclusion: Low-Level laser is effective in inducing reparative dentin formation combined with bioactive EndoSequence Root Repair Material. Both of fast-set and regular-set EndoSequence Root Repair Material produced favorable pulpal responses. Runx2 staining is more intense than osteocalcin and more expressed in Low-Level laser treated group compared to non-lased group.
