Abstract : This study's main target is to investigate the possible role of local honey and Taifi rose oil applications in curing the fungal infection in the vagina. The research also scopes the light on the vaginal microbiome signature of non-pregnant women, its dysbiosis due to the fungal infection, and the impact of the recommended treatment towards restoring microbial homeostasis as a prerequisite. This study includes two parts, in vitro and in vivo. In the first part, different kinds of honey, mixed with or without the Taifi rose oil, were investigated for their influence scored as an anti-candida activity to select the most promising application for the possible cure of vaginitis In vitro. We examined four concentrations (30, 50, 80, and 95%) of three local kinds of honey (Markh, Qatad, and Sider) and compared them with Manuka honey, while two concentrations of the Taifi rose oil (1 and 2%). The second part is in vivo by using the Suggestion treatment from laboratory experience (Markh honey with or without mixing with Taifi rose oil) to treat vaginal infection and its effect on the vaginal microbiome in vivo. The vaginal microbiome was detected in six women with vaginal infection. The study was conducted in comprehensive specialized polyclinics in Jeddah. The participants were randomly divided into honey groups and honey mixes with rose oil groups (n, 3 per group) and were assessed both before and after seven days of treatment. The results were analyzed by next-generation sequencing (NGS) in BGI China. Concluded that honey treatment has a high effect on rebalancing the abundance of Lactobacillus than honey mixed with rose oil, whereas honey mixed with rose oil had a strong effect to lessen the abundance of Candida species. Successful results may indicate the use of the new mixture (Markh honey with or without mix with Taifi rose oil) as a therapeutic drug for patients with vaginal candidiasis.
