Abstract : The alterations of dynamic brain functions in Alzheimer's disease (AD) remain unclear. The present study is designed to determine the protective effect of the Magnesium Threonate (MgT) (296 mg/kg., b.w.) against neural oxidative damage caused by aluminum chloride (AlCl3) (10 mg/kg b.w.) induced rat model of AD in hippocampus rat brain. It was assessed by attenuated total reflection spectroscopy (ATR-IR), biochemical test, and histologically by light microscope. Adult male Albino Wistar rats (N=16) were divided into the Healthy controls group (C), Alzheimer's group (AD), (Mg) group, and the protective group (Port). The overall findings demonstrate the alterations in the significant biochemical constituents, such as lipids and proteins of the brain tissues of rats. ATR-IR results revealed that AlCl3 caused a significant decrease in the lipid chain packing ratio and the degradation of lipids into large amounts of short-chain fatty acid, with the change in olefinic ═CH and CH2 groups, which could be interpreted as a result of lipid peroxidation. The narrowing in the half band width (HBW) of the sνCH2 sub-band around 2852 cm-1 due to Al intoxication which indicates the presence of trans form fatty acids rather than gauch rotomer. AlCl3 alters the secondary structure of proteins by inducing the breaking of hydrogen bonds that stabilize the protein structure. There was a significant decrease in lipid to protein ratio in AD group. AD group showed a significant increase in amyloid precursor protein, tau protein, and malondialdehyde levels with a decrease in the glutathione and superoxide dismutase activity and albumin level. The histological examinations showed clumpy cell neurons or reduced pyramidal cells and scanty neurofibrillary tangle, which indicated neurodegeneration compared to the control group. Biophysical, biochemical, and histologic aberrations were attenuated on treatment with MgT. Conclusions: The study showed that MgT has a protective role in Alzheimer's disease by acting against excitation, leading to neuronal cell death (excitotoxicity), and reduced oxidative stress.
